Actinomycin D intercalates into DNA and thereby inhibits the transcription. It binds preferentially to guanine and blocks the RNA polymerase during the elongation step. A high dose (2ug/mL, 1.59uM) inhibits the transcription of all RNA species, lower concentrations (40ng/mL, 32nM) inhibit the synthesis of rRNA.

Actin, pan Ab-5 Hot! LVMS1295

Highly recommended for monitoring total protein load on Western blots.

Comments: Ab-5 reacts with all six known vertebrate isoforms of actin (M.W. ~42kDa). It also reacts with two cytoplasmic actins (Beta, Gamma) which are highly homologous to one another but which differ from the muscle actins at about 25 amino acid residues.

Applications and Suggested Dilutions:
- Immunoprecipitation (Denatured verified) (Use Protein G) (Ab 2ug/mg protein lysate)
- Western Blotting (0.5ug/mL for 2hrs at RT)
- Immunohistology (formalin/paraffin) (Use at 1:400 for 30 min. [Staining of formalin-fixed, paraffin-embedded tisssues requires boiling tissue sections in 10mM citrate buffer, pH 6.0 for 10-20 min. followed by cooling at RT for 20 min.]

Less toxic alternative to PMSF, pH stable.
AEBSF irreversibly inactivates thrombin and other serine proteases (e. g. chymotrypsin, kallikrein, plasmin, proteinase K, trypsin) by sulfonylation of a functional group in the active center of the enzyme. AEBSF (LD50 oral, mouse 2.8g/kg) is a substantially less toxic substitute for PMSF and DFP.

For the preparation of 1 Liter liquid for LB agar plates weigh 10 g tryptone, 5 g yeast extract, 5 g NaCl and 15 g agar. Add 1 ml 1 N NaOH to the dissolved powder. The medium will be autoclaved for 25 minutes. At a temperature of ~50℃, it is still liquid and additives (e. g. antibiotics) can be added. If 32 to 40 ml LB agar per plate are used, one liter will result in approx. 25 to 30 plates. Freshly poured plates cool down with a slightly opened lid and turn them upside down when stored at +4℃.
